Correlation of ENT1, TK1, dNT, TMPK, and PCNA Expressions with F-18 FLT Uptake in Colorectal Cancer

PURPOSE

The purpose of this study was to examine equilibrative nucleoside transporter-1 (ENT1), thymidine kinase-1 (TK1), deoxynucleotidase (dNT), thymidylate kinase (TMPK), and proliferative cellular nuclear antigen (PCNA) expressions in operatively removed tissue samples by reverse transcriptase-polymerase chain reaction (RT-PCR) to elucidate the mechanism of 3'-deoxy-3'-18F-fluorothymidine (FLT) uptake, in colorectal cancer.

METHOD AND MATERIALS

A total of 13 patients with colorectal cancer were examined with FLT PET. The maximal standardized uptake value (SUV) in colorectal cancer was calculated. The expressions of ENT1, TK1, dNT, TMPK, and PCNA in operatively removed tissue samples (cancer tissue and normal tissue) were compared and they were also compared with FLT SUV.

RESULTS

All colorectal cancer lesions showed focally increased uptake of FLT PET. The mean (±S.D.) FLT SUV in colorectal cancer was 5.66±2.26. The mean values of cancer tissue content of ENT1, TK1, TMPK, and PCNA mRNA expressions in cancer tissues were significantly higher than the corresponding values in normal tissues. There was no significant correlation between FLT SUV and either ENT1 or TK1 or dNT or TMPK or PCNA mRNA expression.

CONCLUSION

In this preliminary study, colorectal cancer was able to visualize on FLT PET. Significant differences in ENT1, TK1, TMPK, and PCNA mRNA expressions between cancer tissues and normal tissues were observed. The expressions of ENT1, TK1, dNT, TMPK, and PCNA were not related to FLT uptake in colorectal cancer.

CLINICAL RELEVANCE/APPLICATION

Colorectal cancer was able to visualize on FLT PET.
